Abstract:

Copy Number Variations (CNVs) of regions of the human genome have been associated with multiple diseases. We present a rigorous mathematical analysis of the CNV problem in a DNA sample using a nanofluidic device, known as the digital array. We show how one can estimate DNA concentration and the associated statistical confidence interval, and then how one can estimate ratio of concentrations of two genes and the associated statistical confidence interval. The latter technique can be implemented as a numerical histogram based algorithm. We also discuss alternative approaches. Finally, we present results of an actual biological experiment on CNV.
